Twenty One Pilots begint in 2009 als groep met Tyler Joseph, Chris Salih en Nick Thomas, die elkaar op school hadden ontmoet. Het drietal brengt in december van dat jaar hun titelloze debuutalbum uit. Als halverwege 2011 Thomas en Salih het wel genoeg vinden gaat Josh Dun verder met Tyler Joseph. Later dat jaar verschijnt het tweede album, Regional At Best. Al het werk betaalde zich in 2012 terug met een platencontract bij Atlantic, waar ze in vrij snel de ep Three Songs uitbrengen. Die zomer touren ze door de V.S. met Neon Trees en Walk The Moon.
Begin 2013 is Vessel hun eerste complete album, waarvan Holding On To You, Guns For Hands en Lovely als single verschijnen.
In het voorjaar van 2015 verschijnt hun album Blurryface, dat later de eerste plaats van de Amerikaanse albumlijst bereikt. Nummers als Fairly Local, Tear In My Heart en Stressed Out vallen in de smaak en met Stressed Out scoren ze in februari 2016 hun Top 40-debuut. Ride wordt drie maanden later de opvolger. Heathens, afkomstig van de soundtrack bij de film Suicide Squad, wordt door Radio 538 uitgeroepen tot Alarmschijf.
Op 5 oktober 2018 verschijnt Trench, het vijfde studioalbum van het duo. Als voorlopers brengen ze de singles Jumpsuit, Nico And The Niners en My Blood uit.
Met Level Of Concern, de track die in het voorjaar van 2020 verschijnt, hoopt het duo steun te kunnen bieden tijdens de coronacrisis die de wereld isoleert. Een deel van de opbrengst komt ten goede aan Crew Nation, een initiatief dat geld inzamelt voor personeel in de muziek- en entertainmentwereld dat wordt getroffen door de maatregelen rond het virus.
