Melissa Jefferson begon haar muzikale loopbaan als rapper en kwam daarna in verschillende groepen terecht, waaronder Lizzo & The Lava Ink en The Shalice. In het najaar van 2013 brengt ze met Lizzobangers haar debuutalbum uit. In 2016 maakt ze de overstap naar Atlantic waar Coconut Oil haar eerste ep is.

Na de release van de single Girls (in 2018) lijkt ze met de opvolger, Juice, met grote stappen de rest van de wereld te kunnen veroveren. Juice staat vier weken in de Top 40. In 2019 krijgt haar volgende single, Truth Hurts, een plekje op de Deluxe-uitvoering van haar derde studioalbum, Cuz I Love You. Die track wordt een nummer 1-hit in de Amerikaanse top 100. Van Good As Hell, dat ondertussen daar ook de top 20 heeft bereikt, verschijnt in oktober een tweede versie waarop ook Ariana Grande te horen is.
Vanaf 25 maart 2022 heeft ze op Amazon Prime haar eigen video-serie, Lizzo's Watch Out for the Big Grrrls, hierin gaat ze op zoek naar danseressen voor haar 2022 tour. Na drie jaar lanceert Lizzo in april 2022 haar nieuwe single About Damn Time wat afkomstig is van haar nog te verschijnen vierde album. In juli 2022 verscheen haar vierde album genaamd Special. Ze brengt twee dagen na de release van haar album de tweede single uit genaamd 2 Be Loved (Am I Ready).
