Een demo van één van zijn eerste nummers, Anyway, komt terecht bij Brian McKnight die het nummer op zijn tweede album plaatst. Het zorgt er voor dat hij als 16-jarige een platencontract kan tekenen bij Interscope.

Inmiddels maakt hij tracks voor acts als Brandy, Jordan Knight, 3T, Mya, Michael Jackson en Christina Aguilera. Als Thicke scoort hij met zijn debuutsingle in 2003 ook meteen zijn eerste hit in ons land met When I Get You Alone. De track, gebaseerd op de vijfde symfonie van Beethoven, komt op de derde plaats. Later dat jaar verschijnt het album A Beautiful World.

Hij tekent een contract met The Neptunes en werkt samen met Pharrell aan het album The Evolution Of Robin Thicke dat in 2006 verschijnt. Van zijn derde studioalbum weet de eerste single, Magic, hem in Nederland weer een hit op te leveren, weliswaar met een bescheiden 28e plaats als resultaat.

De titeltrack van zijn zesde studioalbum is Blurred Lines dat hij samen met T.I. en Pharrell Williams opneemt. De track staat elf weken op nummer 1 en nestelt zich bij de tien grootste hits die ooit in de Top 40 hebben gestaan. Achteraf doet de hit veel stof opwaaien omdat de erven van Marvin Gaye het nummer te veel vinden lijken op Got To Give It Up. De familie eist een vergoeding van ruim 23 miljoen euro, waar de rechtbank in Los Angeles uiteindelijk een boete van bijna 7 miljoen euro oplegt. Eind 2013 speelt hij het nummer samen met Earth Wind & Fire tijdens het Grammy concert dat wordt uitgezonden door CBS.

Het album Paula, dat hij in 2014 aflevert, levert in Nederland geen resultaat. In 2015 werkt Thicke, samen met Verdine White (Earth Wind & Fire) mee aan I Don’t Like It, I Love It van Flo Rida dat door Radio 538 wordt uitgeroepen tot Alarmschijf. In augustus verschijnt zijn samenwerking met Nicki Minaj, Back Together.

In november van 2016 brengt Thicke een nieuwe single uit waarop hij met Juicy J samenwerkt, One Shot.
