Terwijl Donald Glover nog studeerde aan de universiteit van New York begon hij als schrijver voor de NBC-serie 30 Rock waarna hij een rol kreeg in de serie Atlanta. Voor die rol werd hij meerdere keren onderscheiden, onder meer met een Golden Globe.
Op muzikaal vlak brengt hij in 2011 zijn eerste album, Camp, uit als Childish Gambino. Daarna volgen Because The Internet (2013) en Awaken, My Love! (2016). Van dat laatste album kwam zijn grootste succes in de Amerikaanse hitlijst, Redbone, dat in augustus 2017 op nummer 12 wist te komen. Kort voor dat succes kondigde hij aan dat hij er over na zat te denken de naam Childish Gambino niet langer te gebruiken. Van dat idee ziet hij uiteindelijk af en in mei 2018 brengt hij het nummer This Is America uit. De track komt in de Billboard Hot 100 binnen op nummer 1, mede door de bijzondere video van Hiro Murai. De clip is na de release één van de vijf video’s die ooit het snelst over de grens van 100 miljoen views is gegaan.
Inmiddels is Glover een gerespecteerd acteur met rollen in films als The Martian (2015), Spider Man: Homecoming (2017) en Solo: A Star Wars Story (2018).
